Abstract: Las bacterias resistentes y la contaminación hídrica son dos de los problemas más graves que enfrenta la salud pública. En el Ecuador la falta de tratamiento de aguas residuales es una de las principales causas de contaminación, ya que las mismas son dirigidas directamente a los efluentes de agua cercanos. Muchos estudios realizados en los ríos cercanos a la ciudad de Quito muestran altos niveles de contaminación bacteriana resistente a antibióticos, por lo que evaluar nuevas metodologías para la eliminación de este tipo de bacterias presentes en las aguas residuales es de vital importancia para reducir el riesgo tanto a la salud pública como al medio ambiente. Por lo tanto, el propósito del presente trabajo fue evaluar la actividad antimicrobiana del aceite esencial de pino (Pinus radiata), frente a microorganismos resistentes a antibióticos obtenidos de 3 ríos visiblemente contaminados de la ciudad de Quito. El método por el cual se extrajo el aceite esencial fue la destilación por arrastre de vapor ya que muestra ser una técnica sencilla, confiable y de alta calidad. Se requirió 8 kg de materia vegetal para producir 5 ml de aceite esencial en un periodo de 2 meses. Se tomaron muestras de 3 ríos de la ciudad de Quito: Río Machangara, Río San Pedro, Río Guayllabamba. Estas muestras fueron inoculadas en un quimiostato simple de una etapa seguido por una etapa de detección de susceptibilidad a antibióticos, del cual pudimos obtener como resultado microorganismos resistentes a la penicilina. Para comprobar la actividad antimicrobiana del aceite esencial se realizó un protocolo de antibiograma por difusión en disco a 3 dosis distintas: 10ml, 15ml y 20 ml, con un periodo de incubación de 24h a una temperatura de 37ºC. Como resultados se obtuvieron 5 ml de aceite esencial de pino, el cual con las dosis utilizadas en el antibiograma no mostró ningún tipo de actividad antimicrobiana notable frente a las bacterias resistentes a la penicilina. A pesar de que no se obtuvo el resultado deseado se espera que este trabajo promueva la investigación de los aceites esenciales como agentes antimicrobianos en otras condiciones.
Description: The resistant bacteria and hydric contamination are two of the most serious problems that public health faces. In Ecuador the lack of treatment of sewage waters is one of the principal causes of pollution, since these are directly deposited in the near tributaries of water. Many studies done in the rivers near the city of Quito show high levels of bacterial contamination. That is why the purpose of the present thesis was to evaluate the antimicrobial activity of pine essential oil (Pinus radiata), in front of resistant organisms obtained from three rivers visibly polluted in the city of Quito. The method used to extract the pine essential oil was steam distillation since it shows to be an easy technique, reliable and high quality. It was required 8kg of vegetal matter to produce 5ml of pine essential oil in a period of two months. The samples were taken from three rivers in the city of Quito: Río Machángara, Río San Pedro, Río Guayllabamba. These samples were inoculated in a simple one cap chemostat followed by a susceptibility to antibiotics protocol with a treatment of 24 hours, from which we obtained as a result microorganisms resistant to penicillin. To prove the antimicrobial activity from the essential oil a disk diffusion antibiogram protocol was performed in three different concentrations: 10ml, 15ml and 20ml, with a period of incubation of 24 hours, at a temperature of 37ºC. As a result 5ml of pine essential oil were obtained, which with the dosage used in the antibiogram it didn’t showed any type of antimicrobial activity in front of the bacteria resistant to penicillin. Even though the desired result was not obtained it is expected that this thesis promotes the investigation of the essential oils as antimicrobial agents in different conditions.
