Abstract: Resumen: La resistencia a antibióticos es una problemática mundial, el incremento en la prevalencia de estos microorganis-mos tiene gran impacto a nivel económico y especialmente en la salud pública. Estas bacterias resistentes pueden acarrear problemas en el tratamiento de enfermedades causadas por las mismas. Asimismo, a nivel industrial, el mal uso de antibióti-cos en la industria alimentaria ha provocado el esparcimiento de estos microorganismos e incluso su transferencia a otros seres vivos. Los métodos convencionales para la identificación de bacterias resistentes y pruebas de susceptibilidad de estos organismos contra diferentes antibióticos no permiten detectar bacterias con altas tasas específicas de crecimiento y tampoco determinar la resistencia de una comunidad microbiana a diferentes antibioticos. Es por eso que se evaluó un nuevo sistema tecnologico (biorreactor quimiostato de fase simple seguido de un sistema de susceptibilidad a antibióticos) para detectar comunidades bacterianas multiresistentes a antibioticos; este sistema nos permitirá ejercer una presión selectiva para bacterias con altas tasas de crecimiento y además evaluar la susceptibilidad a un antibiotico en una comunidad bacteriana. Se utilizaron alimentos recolectados en mercadados del Distrito Metropolitano de Quito, los cuales fueron inoculados en el quimiostato simple de una etapa para aislar comunidades bacterianas a 3 diferentes tasas de crecimiento. Posteriormente, las muestras aisladas del biorreactor pasaron a una prueba de susceptibilidad a 5 diferentes antibióticos para observar cambio en las densidades ópticas antes y después del tratamiento; seguido, se realizó la secuenciación de tercera generación de las muestras que mostraron poca susceptibilidad a los diferentes antibióticos para identificar los diferentes géneros en cada co-munidad bacteriana y analizar sus antecedentes de resistencia a los diferentes antibioticos. Se encontró que el nuevo sistema tecnológico tiene la capacidad de aislar comunidades bacterianas a altas tasas específicas de crecimiento, seguido del sistema de susceptibilidad en el cual las comunidades bacterianas no mostraron una alta suscep-tibilidad a la gentamicina y ceftriaxona. Además, mediante la idenificación mediante secuenciación, se encontró que el género bacteriano predominante en estas muestras fue Stenotrophomas, cuyos miembros han sido relacionados con agentes patóge-nos y resistencia a antibióticos, por lo que es importante llevar un monitoreo de este tipo de bacterias en alimentos de consumo diario.
Description: Abstract: Antibiotic resistance is a global problem, the rise in the prevalence of resistant microorganisms has a great impact on the industry, especially in the public health area. These multi-resistant bacteria could cause adverse effects in the treatment of disease caused by the resistant bacteria itself. Moreover, in the industry, the overuse of antibiotics in the food industry has caused the spread of these microorganisms and even the transfer of these bacteria from one organism to another. The con-ventional methods used for the identification of resistant bacteria and their susceptibility to different antibiotics do not allow the detection of bacteria with specific high growth rates. In addition, these methods do not allow to determine susceptibility to an antibiotic in a population of bacteria, they are based on pure cultures. In this study, we have evaluated a new methodology (a single-stage chemostat, followed by an antibiotic susceptibility test) to detect bacterial communities multiresistant to antibiotics. This system will allow us to put selective pressure on bacteria with high growth rates and to evaluate the susceptibility to a specific antibiotic in a bacterial community. Food was collected from a popular market in Quito and then inoculated in the bioreactor to isolate microorganisms at three different dilution rates. Afterward, the samples isolated from the bioreactor were tested by a susceptibility test with 5 different antibiotics. This study was based on the fold change of the optical density before and after the treatment. Finally, a massive sequencing of the samples that showed non-susceptibility to the antibiotics used before was performed, which allowed us to identify and analyze the history of multi-resistance of a gender. As a result, the new technological system has de capacity to isolate bacterial communities that grow at high rates. Moreover, the susceptibility test showed that bacteria do not show susceptibility to gentamicin and ceftriaxone. In addition, the identi-fication of the genus by the sequencing showed that the most abundant genus was Stenotrophomas, whose members have been related to some pathogenic strains that are resistant to antibiotics, which is why it is important to have control and monitoring of this type of bacteria in daily consumed food
