Abstract: La brucelosis es una enfermedad causada por bacterias del género Brucella que pueden infectar a especies de animales silvestres o domésticas siendo de importancia a nivel mundial por el impacto económico de la enfermedad en las zonas pecuarias y las infecciones zoonóticas que se convierten en un problema de Salud Pública en países ganaderos y en vías de desarrollo. La enfermedad es endémica en el Ecuador sin embargo no se ha realizado un estudio de variabilidad genética de las especies circulantes mediante métodos filogenéticos moleculares, que complementando con la investigación sobre la prevalencia de Brucella puede proporcionar información importante para encontrar mecanismos para el diseño de estrategias de control de la enfermedad. El objetivo de la investigación fue analizar la información filogenética de tres marcadores moleculares (bcsp31, omp2a y omp2b), e inferir patrones evolutivos para determinar su utilidad en la identificación del género y sus variedades, así como su epidemiología molecular, utilizando un banco de muestras animales y humanas almacenadas en el cepario del Instituto de Investigación en Zoonosis – CIZ de distintas provincias del Ecuador en los años 2008 al 2017. Se aplicó un diagnóstico molecular preeliminar por PCR Bruce-ladder descartando las muestras no conservadas. De un total de 65 muestras válidas 60 correspondían a B. abortus y cinco a B. canis en las que se aplicó una PCR para los genes bcsp31, omp2a y omp2b para luego secuenciar los amplicones. Las secuencias fueron alineadas en una matriz con la cual se realizó análisis filogenético por el software MEGA 10. Se construyeron árboles mediante optimización por Máxima Parsimonia con algoritmo de barridos de ramas para obtener islas de árboles con TBR (Tree Bisection and Reconnection) y remuestreos de matrices con 500 pseudoreplicas de Bootstrap, comparadas con secuencias recogidas de genBank (NCBI). Mediante las topologías se comprobó la capacidad del gen bcsp31 para el diagnóstico del género Brucella por su secuencia altamente conservada, el gen omp2b probablemente sufre una recombinación o alta variabilidad porque no se encontró una estructuración filogenética, mientras que su par omp2a presentó una estructuración filogenética con clados definidos y apoyados acorde con especies filogenéticas que presentan polimorfismos de nucleótidos importantes para establecer patrones evolutivos
Description: Brucellosis is a disease caused by bacteria of the genus Brucella that can infect species of wild or domestic animals, being of worldwide importance due to the economic impact of the disease in livestock areas and zoonotic infections that become a public health problem in developing countries. The disease is endemic in Ecuador, however, a study of genetic variability of the circulating species has not been carried out by means of molecular phylogenetic methods, which complementing with the information on the prevalence of Brucella can provide important information to find mechanisms for the design of strategies of disease control. The objective of the research was to analyze the phylogenetic information of three molecular markers (bcsp31, omp2a and omp2b), and infer evolutionary patterns to determine their usefulness in the identification of the genus and its varieties as well as its molecular epidemiology, using a bank of animal samples and humans stored in the stock of the Zoonosis Research Institute - CIZ from different provinces of Ecuador in the years 2008 to 2017. A preliminary molecular diagnosis was applied by Bruce-ladder PCR, discarding the non-conserved samples. With a total of 65 valid samples, 60 corresponded to B. abortus and five to B. canis in which a PCR was applied for the bcsp31, omp2a and omp2b genes to then sequence the amplicons. The sequences were aligned in a matrix with which phylogenetic analysis was performed with the MEGA 10 software. Trees were built by optimization by Maximum Parsimony with Tree Bisection and Reconnection branch swapping (TBR) and matrix resampling with 500 Bootstrap pseudo-replicates and compared with sequences collected from genBank (NCBI). By means of the topologies, the useful of the bcsp31 gene for the diagnosis of the genus Brucella was verified due to its highly conserved sequence, the omp2b gene probably undergoes recombination or presents high variability because a phylogenetic structuring was not found, while its omp2a pair presented a phylogenetic structuring with defined and supported clades according to phylogenetic species that present important nucleotide polymorphisms to establish evolutionary patterns.
