Título : Divergencia filogenética molecular de las proteínas ACE – 2 (ENZIMA CONVERTIDORA DE ANGIOTENSINA II) Y TMPRSS – 2 (SERIN – PROTEASA TRANSMEMBRANA – 2) EN VERTEBRADOS Y SU RELACIÓN CON EL POTENCIAL DE INFECCIÓN POR SARS – COV – 2

Resumen : enfermedades virales zoonóticas representan una amenaza constante a los sistemas de salud pública mundialmente y, debido a los hábitos expansivos de nuestra economía nos vemos expuestos constantemente a posibles rebrotes o nuevos virus para las que no estamos preparados y, las cuales pueden propagarse descontroladamente debido al estado de híper – globalización en el que vivimos. Como consecuencia, es importante conocer cuáles son los mecanismos de emergencia de estos virus y también, factores que determinan la susceptibilidad de los distintos organismos a la infección de patógenos, así como también su potencial capacidad de transmitirlos a otros organismos y, la relación evolutiva o ecológica entre ellos para establecer posibles estrategias que permitan prevenir la dispersión masiva de dichos patógenos. Se realizaron análisis filogenéticos – moleculares de secuencias de las principales proteínas relacionadas con los mecanismos de invasión celular del virus SARS – CoV – 2 (causante de la pandemia actual Covid-19) provenientes de distintos animales y, se consideraron las relaciones evolutivas con respecto al receptor de ingreso celular del virus con la capacidad de distintos animales vertebrados estar involucrados en el spillover (salto de organismo), portar, replicar y transmitir el virus de forma zoonótica. Mediante análisis bioinformáticos se construyeron matrices de alieneamiento de las secuencias de ACE2-2 y TMPRSS-2 de las clases principales de vertebrados y, se obtuvieron hipótesis (árboles) filogenéticas que permiten visualizar las relaciones evolutivas existentes en torno a las enzimas mencionadas. La topología de los árboles muestra un patrón cercano a la taxonomía y evolución de los organismos y, adicionalmente, sugieren una susceptibilidad decreciente desde el punto más interno del árbol (mamiferos-primates) hacia los clados mas externos (peces). Los primates son, entonces, los vertebrados más susceptibles puesto que sus enzimas son las más similares a aquellas expresadas por los seres humanos, posteriormente los mamíferos carnívoros (cánidos, félidos, mustélidos y pangolines) y quirópteros (murciélagos) que poseen las secuencias de estas enzimas más cercanas evolutivamente y podrían explicar su participación en la transmisión de las enfermedades sin la presencia de cuadros clínicos fatales
Descripción : Zoonotic viral diseases represent a constant threat to public health systems around the world and, due to the expansive habits of our economy we have been continually exposed to possible outbreaks of new viruses for which we are not yet prepared and which could propagate uncontrollably due to the hyper – globalization state we live in. As a consequence, it is important to learn about the emergence mechanisms of these viruses, as well as, the determining factors behind the susceptibility of different organisms to infection from pathogens, their potential capacity for spreading them to other organisms and, the evolutionary and ecological relationship between them in order to stablish strategies that are likely to prevent the massive spreading of said pathogens. Phylogenetic – molecular analysis were carried out using sequences of the main proteins involved in the cellular invasion mechanisms of SARS – CoV – 2 virus that caused the current Covid – 19 pandemic, these sequences belong to different animals and analyzing their evolutionary relationships regarding the cellular entry receptor for the virus compared to the ability of different vertebrates involved in the spillover, carrying, replication and zoonotic transmission of the virus. Through bioinformatic analysis, sequence matrixes were aligned corresponding to ACE – 2 and TMPRSS – 2, taking in consideration the main classes of vertebrates for the obtention of phylogenetic hypothesis (trees) that allow the visualization of existent evolutionary relationships regarding the aforementioned enzymes. The topology of these trees shows a pattern close to taxonomy and evolution of the organisms included and, in addition, they suggest a decreasing susceptibility starting at the innermost point in the trees (Primates – Mammals) towards the outermost clads (fish). Primates are, then, the most susceptible vertebrates due to the similarities shared in their enzymes with the ones expressed by human beings, furthermore, carnivore mammals (canids, felids, mustelids and pangolins) as well as chiropters (bats) who possess evolutionarily closer sequences might explain their participation in the transmission of these diseases without the exhibition of fatal clinical manifestations.
