Abstract: Toxoplasmosis is a reemerging zoonotic disease whose etiological agent is Toxoplasma gondii, an intracellular protozoan parasite, where its definitive reservoir is Felis catus and intermediate hosts such as rodents, birds, wild animals, farm or domestic animals. A diagnostic and molecular ecoepidemiology matrix was generated with the aim of performing a phylogenetic analysis of specific sequences of Toxoplasma gondii that can be used for the rapid molecular identification of any T. gondii isolate in the country and in the region. The optimal molecular marker for the diagnosis of T. gondii was analyzed. The SAG3 marker was found to be suitable for phylogenetic analysis with 170 sequences. MEGAX software was used for phylogenetic analysis by first performing multiple alignment with the CLUSTALW algorithm and manual purification to a size of 566bp. Subsequently, the Maximum Parsimony method with the Tree-Bisection-Regrafting (TBR) algorithm was used to generate the phylogenetic tree. It was observed that SAG3 shows a high genetic variability as it is highly polymorphic, finding a direct relationship between the circulating lineage and virulence, geographic expansion and evolutionary success, which allows its use for diagnosis, as well as in molecular epidemiology and genomic surveillance. In addition, lineage I predominates in the Americas with small introductions in Europe and Asia, while lineage II predominates in Europe and directly affects livestock production and lineage III is found circulating around the world in both definitive and intermediate hosts.
Description: La toxoplasmosis es una enfermedad zoonótica reemergente su agente etiológico es Toxoplasma gondii, un parasito protozoo intracelular, en donde su reservorio definitivo es Felis catus y hospederos intermediarios como roedores, aves, animales salvajes, animales de granja o domésticos. Se generó una matriz de diagnóstico y ecoepidemiología molecular con el objetivo de realizar un análisis filogenético de secuencias específicas de Toxoplasma gondii que pueda ser utilizado para la identificación molecular rápida de cualquier aislado de T. gondii en el país y en la región. Se analizó el marcador molecular óptimo para el diagnóstico de T. gondii. Se encontró que el marcador SAG3 era el adecuado para el análisis filogenético con 170 secuencias. Se utilizó el software MEGAX para el análisis filogenético realizando primero un alineamiento múltiple con el algoritmo de CLUSTALW y una depuración manual hasta tener un tamaño de 566pb. Posteriormente se empleó el método de Maximum Parsimony con el algoritmo Tree-Bisection-Regrafting (TBR) para generar el árbol filogenético. Se observó que SAG3 muestra una alta variabilidad genética al ser altamente polimórfico encontrando una relación directa entre el linaje circulante y la virulencia, expansión geográfica y éxito evolutivo, esto permite su uso tanto para diagnóstico, así como en epidemiología molecular y vigilancia genómica. Además, el linaje I predomina en el continente Americano con pequeñas introducciones en Europa y Asia, mientras que el linaje II predomina en el continente Europeo y afecta directamente a la producción ganadera y el linaje III se encuentra circulando alrededor del mundo tanto en hospederos definitivos como intermediarios.
