Abstract: There is a growing demand for ecosystem biomonitoring which includes the sampling of bioindicator microorganisms. For this, diatoms have been used due to their wide diversity of species and a rapid response to variations in the environment. For their use as bioindicators to be effective, it is necessary to know their biodiversity, and for this purpose morphological identification methods have been used which have presented certain difficulties. As a consequence, molecular identification methods have been implemented through the use of DNA barcodes. The aim of the study is to evaluate the barcodes of the 18s, rbcL and ITS regions in isolated diatoms from different ecosystems of Ecuador. The evaluation was carried out through the construction of phylogenetic trees, which were elaborated using the information available in the NCBI GenBank. Furthermore, the morphological identification was carried out using the original description of the species. The purpose of identification with the 18s and rbcL regions was achieved, presenting rbcL problems in its amplification. The ITS showed a greater divergence compared to the other two regions, but insufficient information was found in the databases. The results showed a greater success by 18s for the identification of the isolated species. However, there is a need for further studies in Ecuadorian species to corroborate the usefulness of this region in identification.
Description: Existe una demanda creciente en el biomonitoreo de ecosistemas el cual incluye el muestreo de microorganismos bioindicadores. Para ello se han empleado a las diatomeas debido a su amplia diversidad de especies y a su rápida respuesta a variaciones en el medio. Para que su uso como bioindicadores sea eficaz, es necesario conocer su biodiversidad, y para este fin se han utilizado métodos de identificación morfológica los cuales han presentado ciertas dificultades. Como consecuencia, se han implementado métodos de identificación molecular mediante el uso de códigos de barras de ADN. El objetivo del estudio es evaluar los códigos de barras de las regiones 18s, rbcL e ITS en diatomeas aisladas de diferentes ecosistemas del Ecuador. La evaluación se la realizó mediante la construcción de árboles filogenéticos, los cuales fueron elaborados usando como base la información disponible en el GenBank de NCBI. Además, se realizó la identificación morfológica utilizando la descripción original de las especies. Se logró el propósito de identificación con las regiones de 18s y rbcL presentando este último problemas en su amplificación. La región ITS presentó una mayor divergencia en comparación con las otras dos regiones, pero no se encontró suficiente información en las bases de datos. Los resultados mostraron un mayor éxito por parte de 18s para la identificación de las especies aisladas. Sin embargo, existe la necesidad de realizar estudios posteriores en especies del Ecuador para corroborar la utilidad de esta región en la identificación.
