Description: The objective of the research was to calibrate for the first time in Ecuador a Water Quality Index (WQI) that explains the behavior of the variables that generate nutrient enrichment and conditions the use of the water resource in the Paute River Hydrographic Basin, at Andean South Region, based on Ministerial Agreement 097-A to determine the uses that water can receive in function of physical, chemical and microbiological parameters. For the sampling the water monitoring records generated by the Hidropaute Business Unit - CELEC EP were taken. during the time series 2012 - 2017 distributed in 11 stations (P1 - P11) along the Paute river that supplies the Mazar and Amaluza reservoirs. For the descriptive analysis and determination of the degree of eutrophy, the PAST statistical program was used; To compare the average values of the variables studied in the time series, the Kruskal-Wallis test was applied (α = 5%); for the classification and ordering of the data, the minimum variance method and the Principal Components Analysis (PCA) were applied, respectively. The classification of water quality was evaluated by applying the (WQI) developed by Brown et al. (1970) for the United States and the ICA calibrated by Moretto et al. (2012) for Brazil, and the calculations were made using the IQAData Software. The results indicated a positively correlated eutrophication gradient with phosphate (r = 0,75) and BOD5 (r = 0,72) where the total phosphate concentration was 0,73 ± 1,51 mgL-1 (Coefficient of Variation, CV = 207,0%), classifying waters of the basin as hypereutrophic. Therefore, the uses attributable to this condition are irrigation and energy production, according to the classification proposed by Thornton & Rast (1994). It was also evidenced that there are significant differences between the average values of the WQI (p<0.05), being lower the average value of the WQI Brazil, a condition that characterizes it as more restrictive, constituting an efficient tool for the monitoring of aquatic ecosystems in the Andean South Region. In addition, in the absence of criteria to qualify the use of water based on total phosphate concentrations, it is concluded that the suggested uses can be incorporated into Ministerial Agreement 097-A, a condition that will contribute to increase the performance of environmental monitoring programs in Ecuador, considering the importance of the phosphate parameter as an environmental indicator of the degree of eutrophication of lacustrine environments.
