Abstract: Debido a las características geográficas y de clima, el Ecuador es considerado un país megadiverso. En la laguna de Limoncocha se ha podido identificar la proliferación de varias especies microalgales, el estudio de estas especies nos permite investigar sobre la biodiversidad y las aplicaciones biotecnológicas que se puedan dar a estas especies. Esta investigación constituye parte del proyecto que tiene por objeto identificar a las microalgas presentes en la laguna de Limoncocha. Se partió de muestras de un consorcio de cianobacterias y microalgas obtenido de la laguna de Limoncocha. Considerando las condiciones óptimas de luz y temperatura para su crecimiento se inoculó en medio sólido Bristol con la técnica de estriado, trascurrido 10 días de crecimiento se tomó una muestra algal y se realizó 7 diluciones seriadas que se inocularon en medio sólido Bristol, observando el cultivo axénico en la dilución 105 y 106. Se realizó una identificación microscópica utilizando claves dicotómicas para identificación de microalgas y cianobacterias con un laboratorio externo, con lo que se confirmó que el cultivo es axénico y corresponde a la microalga Scenedesmus sp. Luego, se realizó cinética de crecimiento en medio de cultivo Bristol y BG11, obteniendo como resultados un mayor crecimiento celular en el medio de cultivo Bristol y con fase exponencial en los días 6, 7, 8 y 9, lo que resultó en una producción de biomasa 0.495g/L. Por último, se realizó análisis de contenido proteico de las biomasas obtenidas en medio de cultivo Bristol y BG11 arrojando resultados 38,13% y 34,58% respectivamente.
Description: Due to geographic and climate characteristics, Ecuador is considered a mega-diverse country. In the Limoncocha lagoon it has been possible to identify the proliferation of several microalgal species, the study of these species allows us to investigate the biodiversity and the biotechnological applications that can be given to these species. This research is part of the project that aims to identify the microalgae present in the Limoncocha lagoon. We started with samples from a consortium of cyanobacteria and microalgae obtained from the Limoncocha lagoon. Considering the optimum conditions of light and temperature for its growth, it was inoculated in Bristol solid medium with the striate technique, after 10 days of growth an algal sample was taken and 7 serial dilutions were made and inoculated on Bristol solid medium, observing the culture axenic in the dilution 10 ^ 5 and 10^ 6. A microscopic identification was made using dichotomous keys for identification of microalgae and cyanobacteria with an external laboratory, which confirmed that the culture is axenic and corresponds to the microalga Scenedesmus sp. Then, growth kinetics was performed in Bristol and BG11 culture media, obtaining as a result a greater cellular growth in the Bristol culture medium and with an exponential phase on days 6, 7, 8 and 9, which resulted in a production of biomass 0.495g / L. Finally, protein content analysis of the biomass obtained in Bristol and BG11 culture medium was performed, yielding 38.13% and 34.58% respectively.
