dc.descriptionEn América del Sur se encuentran las especies T. equiperdum y T. evansi, pertenecientes al subgénero Trypanozoon, que causan tripanosomosis, una enfermedad que ha tenido un gran impacto en el sector pecuario en todo el mundo. A pesar de que, existen reportes de la presencia de tripanosomas en países cercanos al Oriente ecuatoriano, en Ecuador hay pocos estudios correspondientes al subgénero Trypanozoon. Por esta razón, el objetivo de este estudio fue implementar técnicas de detección molecular como la amplificación isotérmica mediada por bucle (LAMP) para la detección de Trypanosoma spp. en animales de interés pecuario, y proporcionar una visión regional de la epidemiología de la tripanosomosis causada por el subgénero Trypanozoon, en la provincia de Orellana, Ecuador. El ensayo LAMP se estandarizó en el laboratorio para la detección de T. evansi, dirigido al gen de la glicoproteína variable de superficie (VSG) del Rode Trypanozoon antigenic type 1.2 (RoTat 1.2 VSG). Este ensayo se basa en la variación de la concentración de Mg2+ durante la reacción LAMP para tener un revelado colorimétrico utilizando azul de hidroxinaftol (HNB), una reacción positiva se indica mediante un cambio de color de violeta a azul cielo. La reacción LAMP se llevó a cabo en un tiempo de 30 minutos, tuvo una sensibilidad analítica de 100 copias de ADN y no tuvo reactividad cruzada con T. equiperdum. Se utilizaron las técnicas LAMP y PCR para la detección de Trypanosoma spp. en 19 muestras de campo provenientes de la parroquia García Moreno, Orellana. La detección del subgénero Trypanozoon se realizó por PCR, por medio de los cebadores ESAG 6/7, obteniendo una prevalencia del 68.42% (13/19), la detección de T. evansi se llevó a cabo por medio de LAMP RoTat 1.2 VSG y para comparar la validez de los resultados se realizó PCR para el mismo gen diana. Estos ensayos revelaron 4 resultados coincidentes, 1 positivo por LAMP y negativo por PCR, y 1 positivo por PCR y negativo por LAMP, obteniendo una prevalencia para ambas técnicas del 26.32% (5/19). Estos resultados indican la presencia del subgénero Trypanozoon en los animales domésticos de la parroquia García Moreno e indican un riesgo potencial para todos los animales presentes en la zona. Por lo cual, se hace necesaria la implementación de estudios epidemiológicos y de diagnóstico que incrementen el nivel de precisión y cantidad de muestras.es
dc.description.abstractIn South America the species T. equiperdum and T. evansi are present, that belong to the subgenus Trypanozoon, which cause trypanosomosis, a disease that has had a great impact on the livestock throughout the world. Although there are reports of the presence of trypanosomes in countries near the Ecuadorian East, in Ecuador there are few studies corresponding to the Trypanozoon subgenus. For this reason, the objective of this study was to implement molecular detection techniques such as loop-mediated isothermal amplification (LAMP) for the detection of Trypanosoma spp. in animals of livestock interest and provide a regional overview of the epidemiology of trypanosomosis caused by the subgenus Trypanozoon, in the province of Orellana, Ecuador. The LAMP assay was standardized in the laboratory for the detection of T. evansi, targeting the variant surface glycoprotein (VSG) gene of Rode Trypanozoon antigenic type 1.2 (RoTat 1.2 VSG). This assay was based on the variation of the Mg2+ concentration during the LAMP reaction to have a colorimetric change using hydroxynaphthol blue (HNB), a positive reaction is indicated by a color change from violet to sky blue. The LAMP reaction was carried out in a time of 30 minutes, had an analytical sensitivity of 100 copies of DNA and no cross-reactivity with T. equiperdum. LAMP and PCR techniques were used for the detection of Trypanosoma spp. in 19 field samples from the García Moreno parish, Orellana. The detection of the Trypanozoon subgenus was carried out by PCR, using the ESAG 6/7 primers, obtaining a prevalence of 68.42% (13/19), the detection of T. evansi was carried out using LAMP RoTat 1.2 VSG and to compare validity of the results, PCR was performed for the same target gene. These tests revealed 4 coincident results, 1 positive for LAMP and negative for PCR, and 1 positive for PCR and negative for LAMP, obtaining a prevalence for both techniques of 26.32% (5/19). These results suggest the presence of subgenus Trypanozoon in the domestic animals of the García Moreno parish and indicate a potential risk for all the animals present in the area. Therefore, it is necessary to implement epidemiological and diagnostic studies that increase the level of precision and number of samples.es
