dc.descriptionLa tripanosomosis del ganado es una enfermedad hemoparasitaria de distribución mundial causada por tres protozoos flagelados del subgénero Trypanozoon (T. brucei, T. evansi, T. equiperdum) y una especie perteneciente al subgénero Dutonella (T. vivax), las cuales tienen un impacto socioeconómico significativo, y limitan la productividad del animal en todo el mundo. En América del Sur, están presentes solamente las especies de T. evansi, T. equiperdum y T. vivax, debido a que T. brucei es una especie endémica del África subsahariana, y su transmisión es a través de la picadura de moscas tsetsé pertenecientes al género Glossina spp. De esta manera, el control de la enfermedad depende en gran medida, de una detección correcta y un tratamiento eficaz para prevenir la propagación de este parásito. Pese a que el Ecuador se encuentra en una región donde la tripanosomosis presenta un carácter endémico y amplia distribución, son pocos los estudios que se han realizado respecto a la situación e información epidemiológica acerca de esta patología. Por lo tanto, el objetivo de este estudio fue determinar la prevalencia de tripanosomosis causada por T. evansi en animales de interés pecuario pertenecientes a la provincia de Sucumbíos, a través de la implementación de técnicas moleculares como la amplificación isotérmica mediada por bucle (LAMP). Para ello, se llevó a cabo la estandarización del ensayo LAMP mediante el uso de un indicador metálico conocido como azul de hidroxinaftol (HNB), debido a la reducción de riesgos de contaminación y la detección de ADN con un alto rendimiento. La detección molecular se realizó mediante las técnicas de PCR y LAMP empleando los primers RoTat 1.2 VSG específicos para T. evansi y adicionalmente en la técnica de PCR se empleó un conjunto de primers ESAG 6/7 específicos para el subgénero Trypanozoon. Posteriormente, se evaluaron 19 muestras de distintos animales de interés pecuario pertenecientes a la región Oriente del Ecuador. La prevalencia general obtenida en la provincia de Sucumbíos mediante PCR ESAG 6/7 y PCR RoTat 1.2 fue del 15.78% y 5.26%, respectivamente. Mientras que con el ensayo LAMP se obtuvo una prevalencia general del 10.52% para T. evansi. La prevalencia encontrada en la provincia de Sucumbíos, indica la posible presencia del agente infeccioso en poblaciones de animales de interés pecuario, lo que representa un riesgo potencial para los animales presentes en el oriente del país. Por lo tanto, se debe destacar la importancia de implementar técnicas sensibles, rápidas y específicas que puedan adaptarse fácilmente a cualquier entorno, y de este modo proporcionar datos preliminares acerca de esta patología en el Ecuador, ya que al obtener una detección adecuada acerca de esta enfermedad se pueden tomar las medidas de control necesarias para la prevención y mitigación de la tripanosomosis.es
dc.description.abstractCattle trypanosomosis is a hemoparasitic disease with worldwide distribution caused by three flagellate protozoa of the subgenus Trypanozoon (T. brucei, T. evansi, T. equiperdum) and one species belonging to the subgenus Dutonella (T. vivax), which have an impact socioeconomically significant, and limit animal productivity worldwide. In South America, only the species of T. evansi, T. equiperdum and T. vivax are present, since T. brucei is an endemic species of sub-Saharan Africa, and its transmission is through the bite of tsetse flies belonging to the genus Glossina spp. In this way, disease control depends largely on correct detection and effective treatment to prevent the spread of this parasite. Despite the fact that Ecuador is located in a region where trypanosomosis is endemic and widely distributed, few studies have been carried out regarding the situation and epidemiological information about this pathology. Therefore, the objective of this study was to determine the prevalence of trypanosomosis caused by T. evansi in animals of livestock interest belonging to the province of Sucumbíos, through the implementation of molecular techniques such as loop-mediated isothermal amplification (LAMP). For this, the standardization of the LAMP assay was carried out by using a metallic indicator known as hydroxynaphthol blue (HNB), due to the reduction of contamination risks and the detection of DNA with a high yield. Molecular detection was performed by PCR and LAMP techniques using the RoTat 1.2 VSG primers specific for T. evansi and additionally in the PCR technique was used a set of ESAG 6/7 primers specific for the subgenus Trypanozoon. Subsequently, 19 samples of different animals of livestock interest belonging to the eastern region of Ecuador were evaluated. The general prevalence obtained in the province of Sucumbíos by PCR ESAG 6/7 and PCR RoTat 1.2 was 15.78% and 5.26%, respectively. While with the LAMP assay a general prevalence of 10.52% was obtained for T. evansi. The prevalence found in the province of Sucumbíos, indicates the possible presence of the infectious agent in populations of animals of livestock interest, which represents a potential risk for the animals present in the east of the country. Therefore, the importance of implementing sensitive, fast and specific techniques that can be easily adapted to any environment should be highlighted, and thus provide preliminary data about this pathology in Ecuador, since by obtaining an adequate detection about this disease, the necessary control measures can be taken for the prevention and mitigation of trypanosomosis.es
