dc.descriptionLas bacterias Klebsiella pneumoniae y Enterococcus faecalis son patógenos responsables de varias enfermedades mortales para el ser humano como la neumonía y endocarditis, y animales importantes en la industria alimenticia como las aves de corral. El tratamiento de primera línea para combatir estas enfermedades han sido los antibióticos como la ampicilina y levofloxacina, sin embargo, debido al uso excesivo y mal manejo de estos medicamentos las bacterias han generado resistencia a estos antibacterianos. Por lo que el desafío al que se enfrenta la ciencia en la actualidad es encontrar nuevas alternativas basadas en principios activos naturales de plantas. Una de las plantas que ha llamado la atención por su contenido de alcaloides β – carbolínicos como la harmina es Banisteriopsis caapi, una de las dos plantas con las que se prepara la bebida indígena tradicional y famosa ayahuasca. Por lo que, el objetivo de este trabajo fue evaluar el efecto antibacteriano de extractos de β – carbolinas obtenidos de la planta B. caapi en K. pneumoniae y E. faecalis a través de la técnica de microdilución en caldo. Primero se caracterizó cualitativa y cuantitativamente los extractos mediante HPLC de fase reversa y después de realizo microdilución en caldo para estimar las IC50 y los porcentajes de inhibición de cada uno de los extractos. Efectivamente se observó un efecto inhibitorio sobre el crecimiento de las dos bacterias, sin embargo, el extracto más potente para K. pneumoniae fue el E9 (15,353 μM) y E8 para E. faecalis (26,50 μM) y el más eficaz para las dos bacterias fue E3. La variante antigua (E3 y E9) presentó un mayor efecto antibacteriano que la nueva. Sin embargo, hacen falta más estudios relativos a los mecanismos que analicen la mutagénesis y el daño estructural en el ADN de las bacterias tratadas con extractos de B. caapi.es
dc.description.abstractThe bacteria Klebsiella pneumoniae and Enterococcus faecalis are pathogens responsible for several deadly human diseases such as pneumonia and endocarditis, and important animals in the food industry such as poultry. The first-line treatment to combat these diseases has been antibiotics such as ampicillin and levofloxacin, however, due to the excessive use and mismanagement of these drugs, bacteria have generated resistance to these antibacterials. Therefore, the challenge facing science today is to find new alternatives based on natural active principles of plants. One of the plants that has attracted attention for its content of β-carboline alkaloids such as harmine is Banisteriopsis caapi, one of the two plants with which the traditional and famous indigenous drink ayahuasca is prepared. Therefore, the objective of this work was to evaluate the antibacterial effect of extracts of β-carbolines obtained from the B. caapi plant in K. pneumoniae and E. faecalis through the broth microdilution technique. First, the extracts were characterized qualitatively and quantitatively by means of reversed phase HPLC and afterwards, microdilution was carried out in broth to estimate the IC50 and the inhibition percentages of each of the extracts. Indeed, an inhibitory effect was observed on the growth of the two bacteria, however, the most potent extract for K. pneumoniae was E9 (15.353 μM) and E8 for E. faecalis (26.50 μM) and the most effective for the two bacteria was E3. The old variant (E3 and E9) presented a greater antibacterial effect than the new one. However, more studies are needed regarding the mechanisms that analyze mutagenesis and structural damage in the DNA of bacteria treated with extracts of B. caapi.es
