Abstract: The constant use of fossil fuels and the environmental impact they produce has sparked interest in new renewable energy sources. There is currently a strong trend towards the development of biofuels from third generation biomass, such as microalgae. In this investigation, the microalgae Desmodesmus armatus was isolated and identified from water samples from the center of the lagoon of the Limoncocha Biological Reserve by means of bibliographic comparisons, molecular identification was carried out by PCR (polymerase chain reaction) and tree construction Phylogenetic using the ITS and 18S regions. A culture of ten days was carried out in photobioreactors in batches of 1L with BG11 medium in which 2.5 g/L of microalgal biomass was obtained; in addition, the elimination of nutrients (NO3–, PO43– and COD) is determined daily. The lipid content was evaluated from wet and dry biomass; and free fatty acids (FFA) with dry biomass, using the organic solvents chloroform: methanol (1:2) [CHCl3: CH3OH (1:2)] and HPLC grade ethyl acetate, the best results were obtained with the solvent mixture CHCl3: CH3OH (1:2), whose percentages of lipids from wet and dry biomass were 6.48% and 21.50%, respectively; while the yield by weight for AGL was 52.80%. With the studies carried out and the values obtained, it is not possible to rule out the use of this microalgae for the potential production of biofuels and it is suggested to carry out more experiments.
Description: El uso constante de los combustibles fósiles y el impacto ambiental que estos producen ha despertado el interés en nuevas fuentes de energía renovables. Existe actualmente una fuerte tendencia hacia el desarrollo de biocombustibles a partir de biomasa de tercera generación, como las microalgas. En esta investigación se aisló e identificó la microalga Desmodesmus armatus a partir de muestras de agua del centro de la laguna de la Reserva Biológica Limoncocha mediante comparaciones bibliográficas, la identificación molecular se realizó mediante PCR (reacción en cadena de la polimerasa) y construcción de árboles filogenéticos usando las regiones ITS y 18S. Se realizó un cultivo de diez días en fotobiorreactores por lotes de 1L con medio BG11 en el que se obtuvo 2,5 g/L de biomasa microalgal; además, se determinó la remoción de nutrientes (NO3–, PO43– y DQO) diariamente. Se evaluó el contenido de lípidos a partir de biomasa húmeda y seca; y ácidos grasos libres (AGL) con biomasa seca, usando los solventes orgánicos cloroformo:metanol (1:2) [CHCl3:CH3OH (1:2)] y acetato de etilo grado HPLC, los mejores resultados se obtuvieron con la mezcla de solventes CHCl3:CH3OH (1:2), cuyos porcentajes de lípidos a partir de biomasa húmeda y seca fueron 6,48% y 21,50%, respectivamente; mientras que el rendimiento en peso para AGL resultó de 52,80%. Con los estudios realizados y los valores obtenidos no es posible descartar el uso de esta microalga para la potencial producción de biocombustibles y se sugiere realizar más experimentos.
